The club have refused to disclose the transfer fees for the two players who were outstanding last season.
"Asante and Smallboy are no more with us. I can confidently tell you they have joined Kotoko but will find a worthy replacement," Lions Team Manager Yashin Laryea said.
"We are also talking to some clubs for some players. We will hopefully come out officially when we resume."
Kotoko have embarked on a massive recruitment drive to augment their squad for their title defence and the 2013 CAF Champions League.
