The bottom-based side will for the first time have the backing of their fans to put the Ghanaians to the sword.
It will be recalled that all football activities in the North African country were suspended due to the Port-Said tragedy which resulted in the death of 74 fans seven months ago.
But the country's Ministry of Interior has agreed to allow fans to attend the match.
Zamalek lost the corresponding fixture 3-2 in Accra back in July after twice taking the lead at the Accra Sports Stadium.
New coach Jorvan Vieira has promised to finish restore the Zamalek's pride despite losing his first match in the 2-1 reverse against TP Mazembe.
This will be Chelsea's second trip to Cairo after they were enveloped with a 4-1 defeat to six-time champions Al Ahly early August.
Chelsea will have the services of left-back Richard Kissi-Boateng but right-back Eric Agyemang remain suspended.
Hans Van der Pluijm's side are third on the league and can enhance their semi-final qualification chances by beating the Whites on the road.
