Sunshine now have seven points from four matches to stay in second place in Group A, while ASO Chlef remain bottom of the standings without any points in the group phase of the continental competition.
The other match of the group will be played in Sousse on Saturday night with Etoile du Sahel home to fellow Tunisians Esperance, who lead the group on nine points.
Etoile are third on the table with four points.
On Friday, Sunshine took the lead in first half stoppage time through Cameroonian midfielder Medrano Tamen.
But immediately on restart of the second half, ASO Chlef restored parity through Biyaga Paul Emile.
The home team were reduced to 10 men when Gharbi Sabri was sent off in the 65th minute.
The Nigerians capitalised on this numerical advantage to score the match winner in the 76th minute through giant striker Ukeyima Akombo.
For the next round of matches, Etoile will welcome ASO Chlef on September 1 and the following day defending champions Esperance will host Sunshine in Tunis.

Very happy for sunshine stars. I believe this will spur them to a greater height
